Fish Sexual Development Test

The fish sexual development test assesses the early life stage effects and potential adverse consequences of putative endocrine-disrupting chemicals such as estrogens, androgens, and steroidogenesis inhibitors on sexual development in fish. In the test, fish were exposed to at least three concentrations of the test substance dissolved in water from newly fertilized eggs to the completion of sexual differentiation approximately 60 days after hatching. Within each treatment level and control group, at least four replicates are recommended. At the end of the test, two core endpoints will be measured for each fish: vitellogenin concentration in head-tail or blood sampling, and the proportion of males, females, sexes and undifferentiated fish by gonadal histology. In fish with genetic sex markers, genetic sex is identified to determine sex reversal in individual fish. The combination of the two core endocrine endpoints, vitellogenin concentration and phenotypic (and possibly genotypic) sex ratio, allows the test to indicate the mode of action of the test chemical.

Endpoints of Fish Sexual Development Test

Methods of the Test

  • Our test is a long-term study of fish approximately 60 days after hatching.
  • Fertilized eggs will be exposed to five different concentrations of chemicals in a flow-through system.
  • Prepare and observe four replicates of 30 eggs until juvenile gonad development.
