Isolation and Cultivation of Marine Microorganisms

Microbiologists, ecologists, agronomists, taxonomists, and evolutionary biologists are increasingly interested in marine microbial natural products (MMNPs). The discovery of MMNPs is mainly attributed to marine microorganisms. 16 of the 20 marine cancer drugs in clinical trials are produced by bacteria. Therefore, isolating and culturing a new marine microorganism may be a rapid method to search for new MMNPs. Most microbes from the environment (>99.9%) do not colonize the nutrient-rich agar media used in the past to isolate marine bacteria. This limited cultural capacity can be attributed to artificial conditions in most media, such as a lack of essential nutrients for growth. Although a variety of molecular methods are available for the analysis of microbial communities, culture-based analysis is far from redundant, as only the isolation of individual bacterial species in pure culture can fully characterize physiological properties and comprehensively assess application potential (e.g., bioactive compounds). Therefore, the development of isolation and culture methods is the premise for studying marine microorganisms.
