Isolation and Purification of Microcystins

Risk Assessment of Marine Biotoxin

Preparative reversed-phase liquid chromatography was successfully used to purify two microcystins (microcystin LR and microcystin LA) from cyanobacterial process waste. Isolation protocols included lyophilized cells by methanol extraction, isolation and concentration by solid-phase extraction, and purification by reversed-phase high-performance liquid chromatography1. Various techniques are currently available for the purification of microcystins. These methods not only enable accurate quantification of microcystins in a sample but also provide detailed information about the toxin variants present. The efficiency of any technique used for microcystin purification will be affected by the method used to extract the toxin from the sample. Another important factor in microcystin purification is the requirement for sample concentration. Most analytical methods are not sensitive enough to detect trace levels of microcystins in water and can be affected by co-eluting compounds, especially in environmental or biological samples. Preconcentration with C18 or immunoaffinity SPE is often necessary for the accurate detection and purification of microcystins in these complex samples.
