Sampling of Marine Microorganism

Many different marine ecosystems and their microbial communities have been identified and studied, ranging from ice-covered polar seas to deep-sea hydrothermal vents. Sampling is one of the most important yet often overlooked aspects of oceanography. The successful isolation of obligate barophilic deep-sea bacterial isolates underscores that pressure is an important determinant of marine microbial zones. Many techniques currently used in microbial oceanography require incubation of seawater samples for varying periods of time. The underlying assumption of these methods is that subsequent incubation conditions do not alter the rate of metabolism or biosynthesis in situ. An equally important issue, especially for post-collection hatch measurements, is the concern for clean sampling techniques. Metal samplers, toxic closure components, or other potentially hazardous materials should be avoided. To ensure that the rates measured during post-collection incubations are representative of those that occur in nature, some precautions must be taken.

  • Sampling of Mud Samples
    At present, there are few dedicated microbial mud collectors. When collecting mud samples, benthic mud collectors are often used. CD BioSciences commonly used mud collectors mainly include box-type mud collectors, multi-tube mud collectors and springs mud picker. The number of microorganisms contained in the mud sample is much higher than that in the upper layer of seawater, and it also includes all the microbial species contained in the water layer above the bottom mud.

    CD BioSciences generally adopts two methods of sampling, overall sampling and tissue sampling. Overall sampling is aimed at biological samples that are small in size and cannot be dissected or easily polluted by dissected sampling. Tissue sampling is a biological sample that is larger than the individual and can be dissected to take part of the tissue for fixed-point analysis.

Storage of Marine Microorganism Samples

After water samples and mud samples were stored, the first thing that happened was a slight decrease in the number of bacteria, followed by a large increase in number and a continuous decrease in species. At CD BioSciences, our researchers suggest that storing seawater and sea mud samples at low temperature can reduce the changes in the types and numbers of microorganisms. Therefore, our researchers immediately analyze the water samples.
