Phytoplankton are present in most aquatic environments (freshwater and marine systems) and SRLs are sometimes requested to detect, enumerate and classify these cells for various purposes. Phytoplankton produce fluorescent photosynthetic pigments that can be detected on most standard flow cytometers. The most common fluorescent pigments in phytoplankton (cyanobacteria and other eukaryotic plankton) are chlorophyll a and b as well as phycoerythrin and phycocyanin (phycobiliproteins). All of these pigments can be excited with blue light, but different pigments produce different emissions. Chlorophyll (a and b) are best detected near the emission maximum (660-670 nm) using a bandpass filter. A standard flow cytometer will use a red filter but is best excited using green light, with a fluorescence emission maximum of around 575nm.
